[Usage and dosage] 1. (1) Enteric amoebiasis, 0.4~0.6g once, 3 times a day, a course of treatment for 7 days; Extra-intestinal amoebiasis was treated with 0.6-0.8g once, 3 times a day for 20 days. (2) Giardiasis, 0.4g per time, 3 times a day, treatment course for 5-10 days. (3) Dracunculiasis was treated with 0.2g each time for 7 days. (4) Marsupiosis, 0.2g once, twice a day, course of treatment for 5 days. (5) Cutaneous leishmaniasis, 0.2g once, 4 times a day, 10 days of treatment. Repeat the course after 10 days. (6) Trichomoniasis, 0.2g once, 4 times a day, course of treatment for 7 days; At the same time, 0.5g suppositories can be placed into the vagina every night for 7-10 days. [Adverse Reactions] 1. The most serious adverse reactions of this product are epileptic seizures and peripheral neuropathy at high doses, which are mainly manifested as acroximal numbness and paresthesia. Persistent peripheral neuropathy may occur in some cases with prolonged use. 2. Other common adverse reactions are: gastrointestinal reactions, the most common such as nausea, loss of appetite, vomiting, diarrhea, abdominal discomfort, abdominal pain, taste change, dry mouth, oral metallic taste, etc.; reversible granulocytopenia; (3) allergic reaction, rash, facial flushing, urticaria, itching, etc.; (4) Central nervous system symptoms, such as headache, dizziness, syncope, paresthesia, limb numbness, ataxia and psychosis, multiple neuritis, etc.; others have fever, vaginal candida infection, cystitis, dysuria, black urine color, etc., most of which are reversible, and recover spontaneously after drug withdrawal.

When the drug concentration was 0.2mg/L, the histolytic amoeba could be killed within 72 hours. This product has powerful killing action of trichomonad, its mechanism is unknown. Metronidazole has a killing effect on anaerobic microorganisms, and the metabolites generated when it is reduced in the human body also have an anti-anaerobic effect, which inhibits the synthesis of deoxyribonucleic acid of bacteria, thus interfering with the growth and reproduction of bacteria, and ultimately causing bacterial death. Carcinogenic to some animals.



[Pharmacokinetics] After oral or rectal administration, it can be rapidly and completely absorbed, and the protein binding rate is less than 5%. After absorption, it is widely distributed in all tissues and body fluids, and can pass the blood-brain barrier. The effective concentration of the drug can appear in saliva, placenta, bile, milk, amniotic fluid, semen, urine, pus and cerebrospinal fluid. Concentrations of the drug in placenta, milk, and bile are reported to be similar to those in blood. The serum concentration of cerebrospinal fluid in healthy subjects was 43% of that in the same period. In a few patients with brain abscess, the concentration of pus (34~45mg/L) was higher than that of blood (11~35mg/L) in the same period after daily administration of 1.2~1.8g. The concentration of drugs in the pus after ear infection was above 8.5mg/L. The serum concentration reached its peak 1~2 hours after oral administration, and the effective concentration could be maintained for 12 hours. After 0.25g, 0.4g, 0.5g and 2g oral administration, the plasma concentrations were 6, 9, 12 and 40mg/L, respectively. 60~80% of this product is excreted through the kidney. About 20% of the original drug is excreted from the urine. The remainder is excreted in the urine in the form of metabolites (25% as glucuronic acid conjugates, 14% as other metabolic conjugates), 10% in the stool, and 14% through the skin.
