[Ingredients] Ranitidine hydrochloride, its chemical name: N '-methyl -N-[2-[[[5-[(dimethylamino) methyl] -2-furanyl] -methyl] thioethyl] -2-nitro-1, 1-ethylene diamine hydrochloride.

[Indications] For the treatment of duodenal ulcer, gastric ulcer, reflux esophagitis, Zollinger-Ellison syndrome and other diseases with high gastric acid secretion.

Usage and Dosage 1. Orally, 150mg (1 capsule) twice a day, or 300mg (2 capsules) once before bedtime.

2. Maintenance: oral 150mg (1 capsule) once nightly.

3. In patients with severe nephropathy, the half-life of ranitidine is prolonged and the dose should be reduced to 75mg (half pill at a time) twice a day.

4. In the treatment of Zollinger-Ellison syndrome, a large dose of 600~1200mg a day (4 ~8 tablets a day) is recommended.

Adverse reactions 1. The common ones are nausea, rash, constipation, fatigue, headache, dizziness, etc. 2. Compared with cimetidine, the adverse effects of renal function, gonadal function and central nervous system were less severe. 3. A small number of patients took the drug after causing mild liver function damage, after the withdrawal of the symptoms disappeared, the liver function also returned to normal. It was suspected that it might be a drug allergic reaction, unrelated to the amount of the drug. 4. Long-term use can reduce the acidity of gastric juice and facilitate the reproduction of bacteria in the stomach, thus reducing the nitrate in the food to nitrite and forming n-nitroso compounds.

When combined with warfarin, lidocaine, diazepam, propranolol and other drugs metabolized through the liver, ranitidine did not increase the blood concentration of ranitidine, resulting in adverse side effects. 2. When used with anticoagulants and antiepileptics, cimetidine is safer than cimetidine. 3. When used with procainamide, the clearance rate of procainamide was decreased. 4. Can reduce liver blood flow, so when used with propranolol, lidocaine and other drugs whose metabolism is greatly affected by liver blood flow, the action of these drugs can be delayed.

1. Pharmacology Ranitidine has a competitive block histamine and H2 receptor binding effect. Inhibition of gastric acid was 5 ~12 times more than cimetidine in mole. Therefore, it is a potent H2 receptor blocker. 2. Toxicology mice oral LD501440~1750mg/Kg. The maximum nontoxic dose was 500mg/Kg for rats (male), 250mg/Kg for rats (female), and 40mg/Kg for dogs for 5 weeks. The maximum nontoxic dose per day was 100mg/Kg for rats and 40mg/Kg for dogs for 26 weeks. No carcinogenic effect was observed in mice by oral administration of 100-200mg /Kg for 114 weeks, and in rats by oral administration of 100-2000mg /Kg for 129 weeks.
